The striker completed an €88 million move to Camp Nou from Liverpool this summer but is yet to play an official match for the Catalans after being banned for four months for biting Giorgio Chiellini during Uruguay's clash with Italy the World Cup.
His suspension ends at midnight on October 25, leaving him free to play against Madrid that same day, and Alves says the former Ajax man is desperate to feature.
"There are a lot of expectations," he told TV3.
"He is a pest and I am sure he is ready. He's working like nobody else and he is really eager to get back to playing."
Alves went on to say that he is unsure what the future holds for him, with his Barca contract set to expire at the end of the season.
"There are just a few months left on my contract," he explained.
"If things work out negatively, that's life. We are in good form, in a good play and with good people to return."
Barcelona face clashes at home to Eibar and Ajax prior to the showdown against Real Madrid on October 25.
